Which 12 gauge ammo is best for home defense?
You put this in the buy sell trade group.

The best is, whatever you have that isn't birdshot. There's a lot of people that like #4 buckshot. I find it easy to find 00 buck pretty much anywhere and it'll get the job done, same for a 7/8ths or 1oz slug.

Well if a shotgun is all you have and don't have the funds to buy an alternative firearm, then I would agree @Downs and running at least a #4 buckshot. Average target distance in a home is going to be less than 10 yds. The pattern at that range is going to be pretty tight even with an 18" barrel.
And Yes, a shotgun will do the job, However, you are typically limited to a max of 8 rds with a massive recoil which makes a follow up shot more challenging and a reload slow.
Honestly, In todays climate, I would highly recommend at a minimum a higher capacity 9mm or 40 cal for quick access to get you to an AR pistol (not an SBR) to put the odds in your favor.
But more than anything, TRAIN, know your weapon well, and practice in your home (unloaded, dryfire drills) and pay attention to where your safe shooting lanes are, especially if you don't live alone!!

Winchester 00 buckshot.

I often have 2 slugs before the buckshot.

My Mossberg JM Pro holds 11 (ghost loaded).

I'll have to double check that number. It's usually stored with a full magazine, ghost load, and empty chamber.

I put the 2 slugs first, so I can rack them out.

I wouldn't alternate them.

12 gauge mossberg with Winchester PDX1 (Three 00 and a slug in each shell) and a Glock 23 (40cal) as a back up.

I like the two rounds of 00 buck, followed by a slug. Kind of punctuates the sentiment of get out and don't get up, er I mean stay out!

Curious, why would you rack the slugs out?
The sound alone will cause them to run off.
Or start unloading in the direction of the sound…
From my time in law enforcement and the military I learned there are 3 types of home invasion.
1. THe people who do not want to see any contact and will run at the sounds of a dog barking/someone walking around the house.
2. The person who is willing to threaten violence against someone but doesnt want to commit violence, much less murder.
3. Those that are expecting violence and are prepared to do whatever they have to, to get away with what they want.

Do not expect persone 1 or 2 to break into your house. If you pull a firearm be prepared to use it, if that is the inevitable outcome, at the same time do not seek a fight, do not waste ammo, do not make noise to give your location or intent. Let robbers get away with material items as those are generally replaceable, your life and your families life is not. Have a plan with the family on where to go/hide and what to do in case of a break in. Everone being alive and safe is the best outcome, the taking of life should only be if all other means have failed.

To answer the OP, a shotgun is a great choice and they actually make home defense rounds such as the winchester rounds mentioned above that have buck/slug combo. Those are meant to do the job and not break through walls. If you have children in other rooms this should be a concern of rounds possibly going through walls (a plan of where to hide helps this as you know where they *should*be). Just because your rounds will not go through walls does not mean the intruders are the same. Also training is extremely important, know the firearm, what the distance is to properly use the ammo you choose and be as comfortable as possible with that firearm. If you are not comfortable with the firearm in controlled conditions you will not use it effectively when you or your families lives are at risk.

Ever see a slug bounce around a forest?

Too much collateral damage from a miss. The slugs are in there for barriers or range. 00 is my go to, the slugs are optional.

Agree. Shotgun in general is a lot of collateral damage.

Advice and collateral damage demonstration from a former SEAL and CIA contractor. Good/funny comments on the shotgun toward the end.
